Asking about work culture sounds like you're someone who is difficult to work with or has high expectations for special treatment.
Job responsibilities is basic stuff.. What's the problem with that one?

I get the preference against discussing salary, but everything else should be on the table. The candidate is interviewing the company just as much as vice versa, so some information before accepting a second interview or offer is required.
I would certainly ask about the job scope and what kind of awesome projects they have to look forward to.
